Pathway 1: Tumor necrosis factor induces the upstream initiator caspases, caspase 8 and 9, Caspase 8 and 9 activate the downstream executioners, caspase 3,6 and 7, then they cleave upwards of 500 key proteins and DNA, which cause the apoptosis. Pathway 2: Tumor necrosis factor induces Mitochondria, which release cytochrome c to bind to apoptotic protease activating factor 1. Then the complex activates pro-caspase 9, which will activate caspase 9 to activate caspase 3 and finally lead the apoptosis.
